In patients with chronic inflammatory bowel disease, there is an abnormal production of especially cytokine and leukotrienes and an increased migration of leukocytes. Mesalazine may have an inhibitory effect on these processes.

Content of packaging Pentasa® Enema.
Pentasa® Enema Suspension is supplied in plastic vials containing 1 gram of mesalazine in 100 ml of liquid. Each vial is individually wrapped in aluminium. The package includes seven plastic bags to insert the vials hygienically.
In the package, the patient receives the following:
- 7 vials (1 gram/100 ml)
- 7 plastic bags

Pentasa® Enema does not require refrigerated storage.
The Pentasa® Enema must be inserted through the anus. To prevent the suspension from prematurely leaving the body through the faeces after administration, it is recommended to have a stool before using the enema.
After the administration, it is advisable to lie down for 5-10 minutes to allow the liquid to penetrate deeper into the colon, and the urge to drain the liquid has disappeared. Therefore, it is recommended to take the enema in the evening immediately before going to sleep.

The dosage of the Pentasa® Enema should be adjusted to the severity of the disease. In general, a once-daily dose of 1 vial of suspension (take the entire contents) is recommended for adults and older children. Use every night at bedtime for two to four weeks. With twice daily dosing, it is possible to take the first dose in the morning and the second dose in the evening. There is little experience and only limited documentation on an effect in children1.
Depending on the individual response to treatment, a doctor may decide in consultation with the patient to combine the use of the enema with other medicines. The tapering schedule below is an example of a combination therapy for IBD in which different drugs are combined.
